說明

糖尿病是本世紀人類最重大的疾病之一。對此疾病的瞭解,大多著重於能量代謝異常中,高血糖及高血脂導致的後續影響,包括對心血管、腎臟、神經系統等之負面作用。而對其生化及細胞層面病理機制研究,則著重在細胞成分的糖化及其引發的生物分子功能損傷及發炎反應,脂肪酸與胰島素抗性的形成,氧化壓力的產生及傷害等等。至今對該疾病較有系統及廣泛的代謝變化研究則乃相當缺乏。

 

背景

代謝症候群為多重危險因子之集合,患有代謝症候群者未來罹患心血管疾病之機率為常人的兩倍,罹患第二型糖尿病之機率為五倍。造成代謝症候群之危險因子包括胰島素抗拒性、血脂異常、肥胖、發炎等。至於前述多重危險因子相互間與不同代謝途徑間的交互作用,及其隨病因進展之變異仍多不明。

 

目的

新興的代謝體學(metabolomics)領域定義為探討低分子量代謝物的全譜型及其變異,以呈現生命體因應生理刺激或基因變異所產生的變化,故特別適用於探討如代謝症候群及糖尿病等具有多重危險因子之人類退化性疾病。

 

方法

在本計劃中,利用代謝體分析(metabonomics)的技術,由代謝物構造及其代謝路徑的鑑定,來獲至對糖尿病代謝異常較完整的了解。
